RESEARCH ACTIVITY

The research interests span the disciplines of organic, bioorganic and medicinal chemistry with an emphasis of study of the interactions between small molecules and their biological (pharmacological) targets. In last ten years, innovative hit and lead compounds for drug discovery were projected and synthesized. These molecules were also used as "chemical genetics" tools to study, in collaboration with biologists and immunologists, some important signal pathways in humans.

The research activity of the lab focused in last year in the design, synthesis and biological characterization of small molecules (mainly cationic and anionic glycolipids) able to modulate the activity of the Toll Like Receptor 4 (TLR4) in the perspective to develop new generations of TLR-directed therapeutics.

1) Synthesis and biological characterization of new molecules active on TLR4 signaling. In collaboration with Jerrold Weiss, (Univ. of Iowa), Francesca Granucci (UNIMIB) Ivan Zanoni (Univ. Molinaro and A. Silipo (UNiv. of Naples), S. Martin-Santamaria (CSIC, Madrid), D. Andreu (PFU, Barcelona), J. Jimenez-Barbero (CIC bioGUNE, Bilbao), R. Jerala (KI Lubljana, Slovenia), T. Huser (UNiv. of Bielefield, Germany), J. Mareque (CIC biomaGUNE, San Sebastian, Spain)

3) Interaction between bacterial LPS and LptC protein that transport LPS from inner to outer membrane in bacteria. In collaboration with Alessandra Polissi and Paola Sperandeo (UNIMIB)

4) New adiuvants based on TLR4-active molecules for anti-allergic vaccines. In collaboration with Lofarma (Milano) and Paola Fusi (UNIMIB)

5) Activity of synthetic TLR4 inhibitors on Amiotrophic Lateral Sclerosys models (ALS), In collaboration with Caterina Bendotti and Massimiliano De Paola (Ist. Mario Negri, Milano)

6) Synthesis of istaroxim analogues as new cardiac drugs (in collaboration with the company CVie Therapeutics, Taiwan and the group of A. Zaza, UNIMIB)

RECENT SUPPORTED RESEARCH GRANTS

2010-2011: Unit coordinator of PRIN grant from Italian Ministry of University and Research (MIUR), title of the project: New multivalent ligands for human galectins as diagnostic and therapeutic agents (3 years grant, 130.000 E for the research unit)

2009-2014: Unit coordinator in the NIH grant AI 59372 "Regulation of MD-2 function and expression", in collaboration with J. Weiss and T. Gioannini (University of Iowa) (5 years, 50.000 E/year )

2010-2013: Unit coordinator in NEDD grant, design on novel pharmaceutically active compounds (80.000E, 2 years).

2013-2016: Unit coordinator of PRIN grant from Italian Ministry of University and Research (MIUR), title of the project: "Italian network for the development of multivalent nanosystems (MULTINANOITA)" (3 years grant, 53.000 E for the research unit)

2015-2018: Coordinator of Horizon 2020-funded MSCA-ETN project TOLLerant "Toll like Receptor 4 activation and function in diseases: an integrated chemical biology approach", Grant Agreement number 642157, consortium of 8 academic and 2 industrial groups, 13 ESR recruited in 2015.

2016-2018: Bilateral project with South Corea founded by Italian Ministry of Foreign Affairs
